would employ when trouble-shooting complaints of localized process slowness?

start with SQL Profiler and Performance Monitor. That's about all I could
tell you without knowing any specifics about your situation...

Usually, most problems lead to 4 usual suspects: CPU, RAM, IO, Poor DB
Design
look at perfmon counters for CPU, RAM and DISK IO to see what is tanking
then go from there.
